As far as I can tell, parallels just makes the interfaces again when it launches... But I think the problem occurs when twonky thinks it should use one of the parallels interfaces instead of the real ones! I've had this problem with Air Mouse (for the iPhone).

I've had a very similar problem, and I found the solution to be removing the network interfaces Parallels had made, under system preferences > network...
