I cannot see anything obviously wrong with the Mediainfo details, except the audio stream is MP3 variable bitrate. Many hardware mediaplayers cannot play variable bitrate MP3 audio. Try re-encoding with constant bit-rate audio

Also on your Win7 machine,
Check the Power Management settings. Make sure they are not set to go into Standby or Hibernate.
Check your Anti-virus and Firewall software and make sure they are not interfering.
PS. There is no log on the WDTV Live that you can access.

Umm, starting to run out of ideas.
The only other possible change you could make to the Yamaha Receiver profile in clients.db is to also add-
DX:WMC
to the profile. Seems some devices need this and it was in the 4.x Yamaha Receiver profile.

If you have a corporate lock down on your PC then you need to go and talk to your IT guys.
At a minimum you need Port 9000 open (default for Twonky) and the firewall must allow multicast broadcasts. Neither are usual in a corporate environment.

Use mediainfo (download here http://mediainfo.sourceforge.net/en) to analyze one or more of your files that do not play and post the TEXT output here. Chances are that there is something in the way the files are encoded that prevents your TV from playing the files.

If the subtitles are embedded in your MKV files then you need to see if your media player supports embedded subtitles when playing from a DLNA server (aka TwonkyServer). Yes that statement is a bit ambiguous. But no, Twonky doesn't stream ISO files and there are no DLNA media players (to my knowledge) that could play them.
DLNA media players want a streamed media file. ISO files are archived directory/file structures so not media stream files.
