It sounds like a problem either with your NAS (4 GB file size limit?) or the files themselves, I'd suspect the first. Running 6.0.30, and older, versions on my Ubuntu server I've never seen that problem.
If you enable the logging, does the Twonky log show anything? If you upgrade to 6.0.30 does the problem go away?

There's lots of information in the forum on enabling transcoding. Check the files ending in .location in cgi-bin inside the Twonky install directory. Edit those and set the full path to the relevant executables (excluding the executable, just the directory) - I think you then have to restart Twonky.

Transcoding is CPU intensive - your NAS may not be able to keep up, but it will depend on the NAS, source media type and the type it's being transcoded to (in particular, the codecs being used).

Rather than creating DVD images, you may want to encode the DVD into MP4 or MKV, or whatever format is supported by your clients. I personally use Handbrake for that step.

I have another of the Sony Blu-ray players that's just been DLNA enabled. I have the same problem playing MKV files, but a Google search shows that people are having problems with a wide range of DLNA servers - not just Twonky.

You need to ensure that you installed twonkymedia in to the directory that script expects it to be in. I seem to recall that it expects it all in /usr/local/twonkymedia.

Samsung's can play MKV files, you just need to ensure you're running 5.1.5 (apparently, not seen it released yet) or 6.0b4 or later.

It seems to work just fine. I ran it with the options "-appdata=/var/twonkymedia-6 -dbdir=/var/twonkymedia-6/db", having created the directories in question. Note that it is up to Beta4 last time I checked.

XBMC uses files in folders called "folder.jpg" and "fanart.jpg" to attach images to folders. See their documentation.
