Have Twonkymedia 5.1.3 running. All is fine (albeit some excessive use of CPU maintaining the library).
Delete 5.1.3 and install 6.0.
Install finishes normally, product fires up, goes to initial config screen via localhost:9000 and system complains it can't find localhost.
Have reinstalled 5.1.3 and all is fine. Have repeated this set of steps and every time, 6.0 will not work as it can't find localhost.

Problem does not occur with 6.0.1 RC5. While the release candidate may have some other quirks (database update still says "in progress ..." even when it appears to be complete) it gets beyond the non-starting install.
Also this is the first time that I have installed a newer level and not had to re-enter license key and directory information. I hope this change continues when the update is officially released

I'm having a similar issue. When I fire up TwonkyServer now I get "Firefox can't establish a connection to the server at localhost:9000". Safari does the same. I haven't heard back from support yet.
UPDATE
So I wanted to try an older version. All I could find was 5.1.6 so I installed it. Same problem. Couldn't connect to localhost. Then I went back and downloaded version 6.0, installed, everything works great. Well so far. I'm still poking around.
