Unable to Access Network Hard Drives

I've been running Twonky Server 4.4.1 on my Synology NAS for many years, but that box is getting old and I've decided to replace it with a home server running VMWare ESCi5.5 and Windows Server 2008 R2.
I have a Windows Server 2008 R2 file server setup and functioning which contains my media collection. Permissions are set so that everybody has read access to this (and as a test I've enables full access to everybody as well, which did not affect my problem).
I have a separate Windows Server 2008 R2 setup to run Twonky Server 8.0.2. This was installed as a service, during the installation.
I have mapped the network drive to the local machine as the E: drive, and this is fully functional.
In the web interface (accessed from my laptop), http://192.168.123.178:9000/webconfig#sharing, if I input the path to the network drive (\\192.168.123.242\Music) I get nothing discovered, if I use the DNS name (\\Mayfield-NA2\Music) I also get nothing. If I click on the browse button, I get presented only with A: and C:, no E:. If I just type in E:, again I get nothing.
Can anybody help, I'm really struggling to get this to work, and don't want to purchase a new licence if this is not going to do what I want.
(I tried Plex Server and this did work, however I was not too impressed that it would not allow me to define my own menu structure for DLNA clients).
