Just got the 4.4b3 up and running on a Mac OS X server.
I specified a directory for Video-Only, Music-Only, Movies-Only.
Content placed in the BASE of those directories works fine, but there's no scanning into subdirectories (as far as I can tell).
Other than specifying each subdirectory as a seperate media location (clunky) is there some way to get Twonky or the PS3 to look into those subdirectories?

My mistake, I was actually at 4.4beta.
I rolled back to 4.3 (as described here:)

Did some experimentation:
4.3 seems to run MP3s and my Pictures (including subdirectories, albums etc), MPEG4 playback is non-existant
4.4 supports MPEG4 playback for some clips I've tried, however the photo subdirectories / albums have broken (music is still fine)
That appears to be the tradeoff currently.
Host is a PowerPC G4 450 running Mac OS X Server 10.4.8
Client is a Sony PS3 60GB running latest firmware
I'll be oscillating between 4.3 and 4.4 depending on whether we're watching movies or browsing photos.
